Abstract

Acute endodontic infections are one of the most frequently treated conditions in endodontic emergency procedures. Microorganisms might reach the connective apical tissues, leading to an intense inflammatory response. If untreated, these infections might progress resulting in life-threatening conditions and systemic complication. Knowing that Streptococcus mutans have been associated with systemic diseases, it will be of extreme importance to detect them in the root canals and dental abscess exudates. Thus, the aims of this study are: 1) To detect S. mutans in samples of infected root canal and their associated acute apical abscesses; 2) To determine the S. mutans serotype in each sample; 3) To detect Cnm glycoprotein of S. mutans in both investigated sites. Patients in need of endodontic therapy due to the presence of pulpal necrosis and associated acute apical abscess (n = 10) were selected. Samples for microbiological analysis were collected from root canals and their associated dental abscesses. DNA will be extracted from the samples from both sites. For the detection of S. mutans in the samples from root canals and dental abscesses, PCR will be performed using the species-specific primers for S. mutans 479F and 479R, described by Chen et al., 2007. The serotype of each sample will be determined by serotype c-, e- or f- specific set of primers according to Shibata et al, 2003, as well as by serotype k-specific set of primers, described by Nakano et al, 2004. S. mutans UA159 (c), B14 (e), OMZ175 (f) and OM88X (k) will be used as reference strains for S. mutans and their serotypes. Samples that show the presence of S. mutans will be evaluated for the presence of Cnm gene by PCR using CNM-541F, CNM-9011R primers.The data will be tabulated and statistically analyzed. (AU)
