Alpha-amylase, acid and alkaline phosphatase in parotid grand from rats treated with sibutramine

Abstract

Sibutramine is a selective inhibitor of neuronal reuptake of noradrenaline and serotonin worldwide used for the treatment of obesity due to its anorectic and thermogenic action. In contrast, many side effects of sibutramine have also been described, especially dry mouth due to its high frequency among patients. Despite the relevance of this problem, it is not known what biochemical changes can occur in the salivary glands of patients taking this anorectic. This work will evaluate the possible effects of sibutramine on the parotid glands of adult Wistar rats (12-16 weeks / 350-450g). The animals will be divided into three groups: treated daily for 28 days with sibutramine at 6 and 10 mg / kg / day per gavage and control group, which received only the vehicle (dimethylsulfoxide and saline). The experimental protocol was approved by the Commission of Ethics in the Use of Animals of the Faculty of Dentistry of Araçatuba - UNESP. After the treatment period, the animals will be weighed and euthanized by exsanguination under general anesthesia. The right and left parotid glands will be removed and their weights measured. Following the glands will be stored at -80 ° C until analyzed. The following biochemical parameters will be evaluated in the glandular homogenate: ±-amylase enzyme activities, acid phosphatase, alkaline phosphatase, tartrate resistant acid phosphatase, and total protein content. Data will be expressed as mean and standard error of the mean (EPM). The normality and homoscedasticity of the data will be analyzed and they will be submitted to the most suitable parametric or non-parametric test. For all tests, the rejection level of the null hypothesis will be set at 5% (p <0.05). (AU)
