Abstract The fluoride ion (F-), when ingested in excessive amounts, can change various cellular functions. Since liver is an organ of high metabolic activity, it has become the target of investigations on the side effects of F, related to alterations in the expression of metabolic proteins, increase in oxidative stress and alterations in the mitochondrial function. Thus, the objective of the present work will be to evaluate the profile of protein expression in the mitochondria of liver of mice chronically exposed to two concentrations of fluoride through the drinking water (15 or 50 ppm), in two experimental periods (20 or 60 days). Thirty-six 21-year-old male Wistar rats will be divided into 6 groups (n = 6 animals per group) according to the concentration of F- to be administered in the drinking water [0 mg / L (control), 15 mgF / L or 50 mgF / L] and the treatment time (20 or 60 days). After the experimental period, the animals will be anesthetized and blood and liver samples will be collected for F- analisys and for proteomic analysis, respectively. F- analysis will be done using the ion sensitive electrode, after hexamethyldisiloxane-facilitated difusion. Mitochondrial extraction will be performed and sample will be prepared for mass spectrometry using FASP protocol. Label-free quantitative proteomic analysis will be carried out (Xevo Q-TOF, Waters). PLGS software will be used to detected alterations in the protein expression between the different groups. Plasma F- data will be analyzed using the appropriate test, after checking for normality and homogeneity (p<0.05).Key words: Proteome, Liver, Mitochondria, Fluoride, Chronic Exposure. (AU) | |
