Abstract

The present study will assess the effects of progestogen supplementation during early gestation on embryo development of pigs. Considering the importance of progesterone in the establishment and maintenance of mammalian gestation as well as its positive influence on the level of energetic substrate available for the embryos in the uterine environment, this study will consist of supplementing gilts with 20mg daily of a progesterone analogue (Altrenogest - Regumate®), orally, in the initial phase of gestation. The females will be divided into 3 groups of 10 animals: CON (untreated females), G1 (females supplemented from 4th to 12th day of gestation), G2 (females supplemented from 6th to 12th day of gestation). The animals will be slaughtered in two distinct periods, 13Ú (15 gilts - 5 per group) and 28Ú (15 gilts - 5 per group) day of gestation (DG), in order to analyze the uterine environment in the period Implantation and embryo-placental development, respectively. In the 13rd DG will be performed quantitative analysis of glucose and embryonic development related proteins present in histotroph as well as analysis of progesterone receptor (PGR) expression in the endometrium. In the 28th DG will be performed the ovarian, uterine and embryonic analyzes. The ovaries will be examined and dissected to determine the volume, weight and number of corpora lutea (CL) and the amount of LH receptors will be measured by RT-qPCR. The number of viable embryos will be analyzed and the development of these embryos will be measured by the measurement of crown to rump, individual embryos weight and embryo spacing. The number of dead embryos will also be evaluated to determine the late mortality rate. The difference between the number of CL and total number of embryos will be considered as early embryonic death. In relation to the uterine evaluation will be analyzed the placental length, length of the embryonic vesicles and placental weight. (AU)
