Association of DNA ploidia and the frequency of micronucle in oral mucosa in chronic smoking patients

Abstract

Tobacco smoking can lead to oral carcinogenesis initiation to development of intra-oral squamous cell carcinoma (SCC). However in this process the initial changes are not visible and the interruption of the carcinogen exposure can prevent the disease's progression. The aim of this study is to investigate the relationship between DNA ploidy and MN frequency in the oral mucosa of chronic smokers. Additionally, a comparison will be made with ex-smokers and non-smokers, to evaluate the cytogenetic changes caused in the lining of the oral cavity and whether these changes decrease with cessation of smoking. Three groups will be evaluated. Group 1: 30 smokers who smoked at least 20 cigarettes per day for 10 years. Group 2: 30 ex-smokers with cessation of smoking for at least 1 year. Group 3: 30 nonsmokers. Exfoliative cytology of tongue and floor of mouth will be performed using cytobrush and the smears will be stained by Feulgen. The presence of MN will be evaluated in 1500 cells, and these data will be correlated with DNA ploidy information provided by the ACIS III equipment. The obtained data will be analyzed statistically by means of (I) calculation of the correlation coefficient between the two variables: Ploidia and MN for G1. Same for G2 and G3; (II) establishment of linear regression lines between Ploidia and MN. Same for G2 and G3; (III) comparison between the angular coefficients of the regression lines, that is, rates of increase of the three groups. Significance level adopted will be the conventional value of 5%. (AU)
