Abstract

Radiotherapy is commonly used for treatment of malignant neoplasms. The bone is widely affected, when the head and neck area is in the radiation field, making it difficult to rehabilitate with implants. Objectives: to verify the viability, differentiation, osteogenic activity and DNA damage of cultured mesenchymal cells from femurs of irradiated rats. Material and methods: Forty rats submitted to radiotherapy simulating a 70 Gy total dose in humans, will receive titanium implants in the right and left femurs. The animals will be divided into two groups: a) Implant Group (Ip) - Control group, with no irradiation, with only implants installed; b) Immediate irradiation group (Ir): Group of implant installation and irradiation in the same day. The animals will be randomly subdivided into 3 groups and will be sacrificed after three days and two weeks of implants installation. Direct fluorescence will be used to verify cell morphology; for cell damage, the micronucleus test; for cell differentiation, MTT [3-4,5-dimethylthiazol bromide] test and for analysis of osteogenic activity tests of total protein content will be used - alkaline phosphatase activity and formation of mineralization nodules. Data will be submitted to statistical analysis and will be correlated with the clinical behavior of the implants. (AU)
