Evaluation of the viability and adhesion of epithelial cells on different materials used in the manufacture of temporary crowns

Abstract

Temporary crowns have as their main function the temporary protection of the remnant of the prepared teeth, restoring the function, aesthetics and stabilizing the occlusion until the installation of a definite fixed crown. Materials used to make temporary crowns should have some properties, such as being easy to handle, biocompatible and non-toxic, ensuring the health of adjacent periodontal tissues and pulp. Thus, the objective of this study will be analyze the adhesion and viability of epithelial cells in contact with the following resinous materials: powder/liquid conventional acrylic resin, bis-acrylic resin and acrylic resin pressed CAD/CAM type, which are widely used for confection restorations and temporary crowns. Standard test specimens will be made (8 mm diameter x 2 mm thick) with the resinous materials, following the manufacturers recommendations, and polypropylene discs will be used as a control. The specimens will be sterilized with ultraviolet light for 20 minutes. Then, these specimens will be placed individually in 24-well plates and on them epithelial cells (NOK) will be seeded (4x105 cells) with complete culture medium. After 24, 48 or 72 h incubation periods of the cells in contact with the test specimens, cell viability analyzes (MTT), cell adhesion and morphology will be performed by scanning electron microscopy (SEM) and fluorescence. The statistical tests to be used in this study will be considered at the pre-determined significance level of 5%. (AU)
