Abstract

The aim of this study is to evaluate the effects of endodontic therapy with different root canal medications in disinfection and return of homeostasis by lipid mediators' quantification in the periapical fluid of teeth with primary endodontic infection. Thirty six single rooted teeth, with primary endodontic infection and apical periodontitis will be selected and randomly assigned into 3 groups (n=12): G1: saline + Ca(OH)2, G2: chlorhexidine, G3: chlorhexidine + Ca(OH)2. Microbial and endotoxic samples will be collected from the root canal, with sterile and apyrogenic paper point, to observe the microbial activity of remaining microorganisms in the root canal by colony-forming units (CFUs) and endotoxin quantification (EU/mL) by Limulus Amebocyte Lysate Assay - LAL, respectively. The intersticial fluid will be collected with sterile and apyrogenic paper points and used for lipid mediators (RvE1, RvD2, LxA4) quantification by Enzyme Linked ImmunonoSorbent Assay (ELISA). The root canals will be instrumented with reciprocating file (WaveOne Gold), a single reciprocating file selected in accord to diameter of root canal and the irrigating solution will be 2.5% NaOCl. After PQM the teeth will be filled with the intracanal medications according to the experimental groups. Fourteen days later, the canal will be re-instrumented and new collections will be performed. Results will be analyzed and submitted to statistical analysis (P <0.05). (AU)
