Antimicrobial activity and physicochemical properties of different pastes used as intracanal medication

Abstract

In necropulpectomies, intracanal medication is recommended to combat microorganisms that may have survived the biomechanical preparation of the root canal system. Therefore, the use of drugs associated with calcium hydroxide in order to increase its antiseptic capacity is suggested. This study will evaluate pH, calcium release, solubility and antimicrobial action on dentin contaminated by oral biofilms, calcium hydroxide + Otosporin, Calcium hydroxide + Nimesulide and Calen paste. Materials and Methods: All experiments will have 3 groups related to the medications in question. PH measurement and calcium release: 10 acrylic teeth in each group will be filled with the folders in question, have the crowns sealed and immersed in deionized water, and after periods of 3, 7, 15 and 30 days will be changed from bottle. In the water in which they were immersed will be measured the pH by means of a pH meter and the calcium released by means of equipped atomic absorption spectrophotometer. Solubility test: In order to evaluate the solubility, the volume of the slides inserted into standard wells in acrylic teeth (N = 10) will be measured initially and after 7, 15 and 30 days of submersion in deionized water using Micro- CT. Antimicrobial action: In vitro biofilm of Enterococcus faecalis will be induced for 21 days on 20 blocks of bovine teeth. Afterwards, the samples will be medicated with the folders in question for 7 days. Through confocal microscopy then images will be captured from the samples and the percentage of live cells will be measured by the bioimage program. (AU)
