Abstract

Recently, procedures involving "Acquired Pellicle Engineering" have been proposed as a new strategy for the control of dental erosion. The incorporation of actives into mouthwash solutions or gels could improve the ability of Acquired Enamel Pellicle (AEP) to protect against erosion. Preliminary experiments from our group revealed that CaneCPI-5, a sugarcane protein, was able to protect the enamel against initial erosion in vitro. Thus, this project presents two general objectives: 1) to compare the protein composition of AEP formed in vitro, in situ and in vivo; 2) evaluate the protective potential of AEP modification with CaneCPI-5 (in different application vehicles) against dental erosion in situ. In the first study, 96 specimens of human enamel (4x4mm) will be made and divided into in vitro and in situ steps. For the in vitro study, unstimulated saliva will be collected from 8 volunteers for the formation of AEP for two hours. The AEP will be collected with boiled cotton balls for 1 minute in SDS solution. For the in situ stage, 8 volunteers will use a removable mandibular appliance for two hours, containing 6 blocks of enamel each. The pellicle will be collected with wick filters pre-soaked in 3% citric acid. For in vivo study, the collection will as described before, but the AEP will be collected directly from vestibular surface of the upper and lower teeth. All samples will be processed for analysis by nLC-ESI-MS/MS. The obtained MS/MS spectra will be compared with human protein databases. The significance of the relative ratios expression will be expressed as p<0.05 for down-regulated proteins and 1-p>0.95 for up-regulated proteins. In the second stage of the project, 360 specimens of bovine enamel will be divided into two large groups: solution (n=180) and gel (n=180); each group will be divided into two types of procedure: erosion (n = 90) and erosion + abrasion (n = 90) and for each of them the study will be conducted in 3 phases according to the treatment: experimental solution CaneCPI-5 0,1 mg/ml (n=30); commercial solution with SnCl2/NaF/AmF (n=30); placebo solution (distilled water) (n=30). 15 subjects will use a palatal appliance for 5 days in each phase, being performed erosive challenge to 0.1%, followed or not by abrasive challenge. The evaluation of wear will be done by contact profilometry. The data will be checked for normality and homogeneity, for selection of the appropriate statistical test (p<0,05). (AU)
