Study of the relation between alveolar bone strain and beta-catenin expression in rats under dental loss

Abstract

To analyze the relationship between Strain's patterns (bone deformation) and beta-catenin expression levels in the occlusal overload environment caused by dental loss in adult rats. MATERIALS AND METHODS: Twenty-four male rats (Rattus norvegicus albinus), Wistar line, at 2 months of age, will be used. At the end of 2 months of age, the upper central incisor tooth will be extracted (right side), and euthanasia will occur in periods 5 (n = 6), 7 (n = 6) and 14 (n = 6) days after Day 0 of the exodontia. In the control group (n = 6), the dentition will be maintained without dental extraction. Euthanasia will occur within 14 days after day 0 of the exodontia. One head per group will be submitted to computerized microtomography. After the three-dimensional reconstruction of the skull and mandible of the rat in each group, the computational simulation for finite elements analysis will be performed to simulate a bite in the incisors. In finite element analysis, the strain patterns will be evaluated after the application of bite force. After euthanasia, the rats of all groups will have their left jaw removed and separated in half. The pieces will undergo routine histological processing and then the immunohistochemical marking will be performed to label expression of the primary beta-catenin antibody, which will be evaluated semi quantitatively (ordinal qualitative analysis) by assigning different scores, according to the number of cells immunolabelled. The results obtained in Strain analyzes will be related to the analysis of beta-catenin expression, proposed in this study. EXPECTED RESULTS: Mechanical and beta-catenin expression of alveolar bone is expected to show possible changes induced by dental extraction induction. (AU)
