Effect of Osteoporosis on mechanical properties of alveolar bone in Wistar rats submitted to dental extraction and diet modification: in vivo, in vitro and in silico analysis

Abstract

To evaluate the influence of Osteoporosis on the anisotropic mechanical properties and microarchitecture of the alveolar bone of Wistar rats submitted to dental extraction and diet modification. Material and methods: 80 female Wistar rats, 2 months old, will be used. The rats will be randomly divided into groups. Ovariectomy will be performed in the rats of the OVX group and surgical simulation in the rats of the SHAM group. Followed 30 days after the day of surgery, extraction of the upper right incisor will be performed and a soft diet will be introduced for half of the animals. The measurement of the bite force on the incisors and the obtainment of weight will be performed daily. The sample will be euthanized on the 30th and 60th days after extraction. After euthanasia, the alveolar bone of the left maxilla and the right alveolus of each animal will be dissected and removed to be subjected to in vitro analysis. The maxillas will be subjected to computerized microtomography to analyze bone microarchitecture and finite element analysis. For the analysis by nanoindentation, the thickness of each bone specimen obtained will be evaluated and will be the mechanical properties of the alveolar bone. Atomic force microscopy will be performed, which will provide a three-dimensional image of the surface at the atomic and/or molecular level, allowing the measurement of various forces at the nanoscale. Expected results: it is expected to elucidate and describe the anisotropic mechanical properties and microarchitecture of the alveolar bone in view of the condition of tooth loss added to the type of diet and Osteoporosis, since they are systemic and local conditions of bone tissue.
