Reproductive, biometric and hematological variables in Danio rerio larvae fed egg yolks from three bird species and challenged with Aeromonas hydrophila

Abstract

Zebrafish, Danio rerio has emerged as a significant model experimental for the study of metabolic diseases. Zebrafish production requires improving feeding protocols for larvae. The objective of this work is to evaluate four feeding protocols with three types of eggs (laying hen, guinea fowl and common quail) and Nauplii of artemia (control group) over the development and immunity of D. rerio larvae. Three hundred and sixty larvae (after 3 days of hatching) will be distributed in 12 aquariums of 15 L (triplicate), water temperature will be kept at 26º C. After 30 days of feeding, fish will be measured and weighed. Half of each group (15 fish) will be challenged by immersion in a Aeromonas hydrophila suspension (5 x 106 CFU/L of water) for one hour. After 6, 12 and 24 hours, animals will be euthanized by deepening the anesthetic plane to collect tissue samples for histopathological and microbiological examinations. The other half of each group will be used to avaliate reproductive variables, after the same feeding period mentioned above, fish will be fed commercial feed for 3 months until they reached their reproductive stage. From this moment, 30 spawns for each group will be evaluated for number of eggs per spawning, number of viable eggs, hatching rate and number of healthy and deformed larvae. (AU)
