Effect of scanning and analysis using micro-CT on evaluation of the properties of repair cements

Abstract

The aim of this study will be to evaluate the influence of different scanning parameters and analysis softwares, using microcomputed tomography (micro-CT), for evaluating the properties of repair materials. Maxillary premolars with two separate roots will be used. Following root-end resection and apical preparation, the teeth will be divided into three groups according to the following root-end filling materials: White MTA, Biodentine and IRM (n=6). The material/dentine interface, volumetric change and porosity will be evaluated after scanning by micro-CT (SkyScan 1272, BrukermicroCT, Kontich, Belgium). Scanning will be performed at 5 µm, after the setting time of the materials, and at time intervals of 7 and 30 days after immersion in PBS. The volumetric change will be evaluated by means of comparison between the initial volume values and those obtained after the time intervals of immersion, in mm³. The porosity of materials will be analysed in the different time intervals. The material/dentine interface will be compared in the different time intervals with regard to adaptation and failures of the materials, as well as changes in the mineralization of dentin walls when bioactive cements are used. These analyses will first be performed with use of the specific softwares of the Sky Scan microtomograph (Bruker-microCT, Kontich, Belgium): NRecon, DataViewer, CTAn and CTVol. The images will be analysed at the resolution of 5 µm and from new datasets they will also be evaluated at 10 and 20 µm for comparison with the analyses between the different resolutions. For each property analysed, the resolution that does not present difference in comparison with 5 µm may be considered adequate for evaluating that specific property. Through a partnership proposed with the School of Medicine, KU Leuven, Belgium, other softwares will also be proposed for each stage of the evaluations, using high quality analysis tools, such as Amira software, Mevslab, Mimics, Trimatic and Proplan. These softwares will be used for comparison with the results obtained in the first stage of evaluation, with use of the previously mentioned softwares. Thus, the best analysis software will be defined, as well as the minimum resolution necessary for reliable evaluation of each property. The data will be submitted to ANOVA and the Tukey tests, with 5% significance. (AU)
