Abstract

Objective: To validate the results of cytokine concentrations from serum and uterine fluid of mares that accumulate uterine fluid (non-experimental cases) measured by the Cytometric bead array kit for human cytokines. Animals: serum and uterine fluid from 12 Quarter Horse mares. Material and methods: Serum and uterine fluid samples from normal mares and mares that accumulate uterine fluid (non-experimental cases) will be quantified at the beginning of estrus (day 1) and on days 2, 3, 4 and 5 after the onset of the estrus by a human Th1/Th2/Th17 CBA kit (BD Biosciences). The readings will be repeated and plotted against a standard curve with recombinant equine Th1/Th2/Th17 antibodies. Data from the two readings will be compared. Clinical Relevance: The CBA assay combines the high specificity and sensitivity of the sandwich ELISA with the versatility of flow cytometry, allowing multiple immunoassays to be performed simultaneously on the surface of small microspheres. This allows the determination of multiple concentrations of cytokines in a single small amount of sample, costing less than the ELISA test. (AU)
