Abstract

The acquired pellicle is considered an organic layer, free of bacteria, formed as a result of the selective adsorption of salivary proteins to the surface of the dental enamel. It is expected that a difference exists in the protein profile of the acquired pellicle formed in situ or in vivo, which has never been evaluated so far. Thus, the objective of this project will be to compare the protein composition of acquired enamel pellicle (AEP) formed in situ and in vivo. In the study, 48 specimens of human enamel (4x4mm) will be made for the in situ stage, 8 volunteers will be molded with alginate and will use a removable mandibular device (Bauru in situ Pellicle Model - BISPM) for two hours, containing 6 blocks of enamel, 3 for each side. The pellicle will be collected with wick filters pre-soaked in 3% citric acid, then will be scrubbed on the specimens and frozen until the preparation of the proteomic analysis. In the in vivo stage, the same volunteers will be used and the collection will be in the same way as the in situ stage, however the AEP will be collected directly from vestibular surface of the upper and lower teeth, after meticulous prophylaxis. All samples will be processed for analysis by nLC-ESI-MS/MS. The obtained MS/MS spectra will be compared with human protein databases. The significance of the relative ratios expression will be expressed as p<0.05 for down-regulated proteins and 1-p>0.95 for up-regulated proteins. (AU)
