Effects of proanthocyanidins on modulation of inflammatory response of gingival fibroblasts

Abstract

Development of periodontal disease is directly related to expression and activity of metaloproteinases (MMPs)by oral mucosa cells. Superexpression of these enzymes leads to higher tissue degradation resulting in accelerated progression of periodontal distruction. Therefore, the application of agents that may modulate synthesis and activity of MMPs could be a feasible therapeutic approach to periodontal disease. Proanthocyanidins are natural phenolic components of seeds and fruits that act like cross-linkers and inhibit MMP. The aim of this study will be to evaluate the effect of grape seed proanthocyanidin (PA) on modulation of oxidative and inflammatory response by gingival fibroblasts. Fibroblasts will be seeded in 96 well plates and exposed to inflammatory stimulus (TNF-alpha), followed by PA at 0,5 and 1 in serum-free culture medium (DMEM). After 24 hours the effects of this treatment on oxidative response (oxygen reactive species) and synthesis of interleukin-6 and metalopproteinase-9 (ELISA) as also gelatiolytic activity (in situ zymography) will be determined. Data will be analyzed by specific statistical tests considering 5 significance level. (AU)
