Abstract

Endodontic cements are materials that influence the periapical tissues and cause tissue reactions of characteristics and intensities depending on the type of cement used. Supplementation with substances such as omega-3 and melatonin have proved to be excellent anti-inflammatory agents and inducers of mineralization. The aim of this study was to evaluate the influence of omega-3 and melatonin supplementation on the tissue response of endodontic cements using the ISO 10993-6: 2007 model for subcutaneous tissue implantation tests. Seventy-Two Wistar rats were divided into three groups with eight animals in each group. The study will have the folowing groups: control rats (C), omega-3 (O) and mice supplemented with melatonin (M). In each rat, 4 polyethylene tubes will be implanted, 1 empty tube for control, and the remaining ones containing one of the types of endodontic cements selected for the study (Sealapexè, AH Plusè and Endofilè). The evaluation periods will be 5, 15 and 30 days (n = 8 / time). After sacrifice, the tubes and surrounding tissue will be removed and processed for histological and immunohistochemical analysis. Inflammation will be assessed in H.E-stained tissue sections and by immunohistochemical labeling for interleukin 6 (IL-6) and tumor necrosis factor alpha (TNF-a); the collagen, through the coloring of Picrosírios red; the biomineralization, by means of the Von Kossa coloration and in cuts without staining by polarized light. The statistic will be performed according to each type of analysis and the significance level of 5% (p <0.05) will be considered. (AU)
