Cell viability evaluation of pulp capping membranes with Portland cement

Abstract

Ideally, dental products should have bioactive, biodegradability, and non-toxicity properties always aiming at different applications. Among pulp capping materials, Portland Cement (PC) is a compatible and low-cost alternative. The PC association with new drug-delivery systems, such as chitosan, is a promising field for the development of materials and research. This study aims to develop and formulate membranes for dentin-pulp biostimulation containing PC and evaluate the cell viability in cell culture of fibroblast from deciduous teeth (DHPF). The PC-based chitosan membranes will be developed. PC will be incorporated in the bandages, as follows: pure and associated with radiopacifiers (zirconia and iodoform). The cell viability assays will use DHPF previously cultured, frozen, and stored at the biorepository of the School of Dentistry of Bauru (University of Sao Paulo). To assess cell viability, Crystal Violet assay will be performed at 24, 48, and 72 hours after the contact of DHPF with the culture medium conditioned with the membranes. The obtained data will be submitted to statistical analysis. The level of significance of 5% will be adopted to consider the differences statistically significant. (AU)
