Evaluation of the healing action of topical Ozonotherapy in surgically induced cutaneous wounds in Wistar rats

Abstract

Ozone therapy is an oxidative therapy that generates oxidative stress, and consequently antioxidant and healing effect on the body. Since studies in more specific and controlled environments on topical ozoniotherapy are scarce in the literature, the present study aims to evaluate the cicatrizing action of ozoniotherapy with ozonated sunflower oil and insufflation of ozone gas in bags, isolated or associated, in surgically induced skin wounds in Wistar rats. It will be used 120 healthy rats of the Rattus novergicus species, males, Wistar male, 16 weeks old, and body mass between 350 and 450 grams. The rats will be randomly assigned to five groups of 24 animals: Group 1 - rats with wounds submitted to treatment with saline solution (0.9%); Group 2 - rats with wounds treated with sunflower oil; Group 3 - rats with wounds treated with ozonated sunflower oil; Group 4 - rats with wounds submitted to treatment with insufflation of ozone gas in bags; Group 5 - rats with wounds submitted to the combination treatment of ozonated sunflower oil and insufflation of ozone gas in bags. Twelve rats from each group will undergo euthanasia at two different times: seven days after starting treatment and fourteen days after the start of treatment, for electrocardiographic evaluation; wounds tomography evaluation; haematology evaluation; renal and hepatic function serum biochemical evaluation, and urinalysis; wounds, kidney and liver histological evaluation; and evaluation of scar tissue tension. The macroscopic evaluation will be daily. Statistical analysis will be used for analysis of variance (ANOVA) for repeated samples, with contrasts by Tukey test and Kolmogorov-Smirnov test. Data that do not assemble the normality characteristics will be used Friedman test. For comparison between the four groups of the study, analysis of variance for independent samples (ANOVA One way) for normal data, and Kruskal-Wallis test, for data outside the normality conditions will be used. Differences will be considered significant with p  0.05.
