Abstract

The first cell differentiation event in mammals consists in the segregation between the inner cell mass (ICM) and the trophectoderm (TE). In mice, HIPPO signaling pathway plays an important role in the control of genes that define ICM/TE segregation. In bovine, there is evidence that the same genes do not participate in a similar manner, but HIPPO pathway may still contribute to this biological event. The present project aims to test the hypothesis that LATS1/2 activity is required for ICM differentiation in bovine embryos. We intend to eliminate LATS through gene editing using CRISPR/Cas9 in in vitro produced embryos. Embryos will be cultured for assessment of blastocyst formation rates, cell distribution, gene and protein expression. We expect to inhibit ICM due to absence of LATS-mediated HIPPO signaling. Consequently, this project would provide a better comprehension in the mechanisms of ICM/TE segregation, which can be useful to understand failures in the vitro production processes. (AU)
