Abstract

The resistance to third-generation cephalosporins (3GC) is an important public health concern worldwide since they are widely used drugs to treat human infections. Several studies report the transmission of bacteria and resistance genes from food to humans via handling or consumption. Detection of Escherichia coli in food indicates possible fecal contamination, and the presence of 3GC-resistant strains points out the possibility of transmission of resistance genes to the consumers' gut microbiota. Research in antimicrobial resistant bacteria from feedlot-lambs is a neglected field in Brazil, although internal consumption of lamb meat shows an exponential increasing in the country. Thus, this project intends to analyze the presence of 3GC-resistant E. coli in lamb meat. For this, 50 retail lamb meat samples will be acquired in São José do Rio Preto, SP. Twenty-five grams will be put in 225 mL of peptone water supplemented with 4 mg/L of ceftiofur and incubated at 37 °C for 20 hours. The resulted broth will be inoculated in MacConkey agar supplemented with 4 mg/L of ceftiofur. Up to 100 presumptive E. coli colonies will be inoculated in a new agar and then preserved. The isolates will be identified by automated system and submitted to antimicrobial susceptibility test by disk-diffusion. The 3GC-resistance determinants will be investigated by PCR and then sequenced. Isolates will be screened by ERIC-PCR and typed by MLST.
