Identification of genes codifying ESBLs, quinolone resistance and virulence in E. coli isoltaed from chickens, chicken meat and patients with urinary tract infections

Abstract

The urinary tract infection (UTI) by uropathogenic Escherichia coli (UPEC) resistant to beta-lactams and quinolones are an important public health problem, because these are the main recommended antibiotics and treatment failure can result in evolution for severe renal failure, sepsis and death. The main reservoir is UPEC the gastrointestinal (GI) tract, and infection occurs after the migration of the bacteria to the perianal region and subsequently into the urethra. In recent years, ITU has been considered as zoonoses and food-borne diseases, based on evidence that meat, especially chicken meat, are a vehicle for the transmission of UPEC from livestock to humans. Brazil is the largest producer of poultry meat in the world, and so, for reasons of public health and business, it is important to know the profile of virulence and resistance in E. coli isolated from chickens and chicken meat produced in the country. In a previous study by our group, E. coli harboring the genes blaCTX-M-2 and qnrB-like that codify resistance to extended spectrum beta-lactamas and decreased susceptibility to quinolones were isolated from samples of chicken meat. In this project, we propose to conduct concurrent evaluation of E. coli resistant to third generation cephalosporins and quinolones isolated from chicken meat, broiler chickens and patients with UTI. The profile of antimicrobial susceptibility genes responsible for ESBL production and resistance to quinolones, and the diversity of virulence genes will be investigated and compared. These bacteria will also be compared in terms of phylogenetic group and genetic similarity by PFGE and multilocus sequence typing (MLST). The results of this study will contribute to knowledge about the spread of resistance genes in UPEC and the role of the production chain of poultry meat in the spread of antimicrobial-resistant UPEC, and in future may be used for the development of food safety programs for the Brazilian meat productive chain.
