Glycemic and oxidative stress profile of rats exposed to intrauterine diabetes and submitted to the hyperlipidic diet in the post-natal period

Abstract

Changes occur during intrauterine life that are capable of influencing both the embryonic and fetal development and the adult life of the offspring, increasing the risk for the diabetes onset. Among these diseases, Diabetes mellitus may be cited, which promotes an unfavorable intrauterine environment. The understanding of existing mechanisms regarding the tolerance of the adaptive responses of the organism is unclear. There is evidence that diseases manifest themselves when the nutrients in the environment after birth are considerably different from those to which the fetus was exposed during the intrauterine environment. Thus, if there is a parallel between the current human situation in which children of diabetic mothers can be exposed to inadequate diet in postnatal life, the objective of this study will be to evaluate how the fusion of these two unfavorable conditions (intrauterine and post- natal) will influence the glycemic and oxidative stress profile of the rats. To obtain the parental generation, the rats of the Sprague dawley strain received subcutaneous injection of Streptozotocin (70 mg / kg) on the 5th postnatal day (this experimental part was previously performed in the doctoral project of one of the students of the laboratory, which will be monitored in the rest of the experiment - PhD work in partnership with this project of Scientific Initiation). The diabetes will be confirmed with the oral glucose tolerance test (OGTT) at the 75th day of life. At 90 days of life, the rats will be mated to obtain the offspring that, after weaning, will be distributed in the experimental groups: Fcont S - offspring of control mothers fed standard diet, Fcont HL - offspring of control mothers fed with hyperlipidic diet, Fdmod DP - offspring of diabetic mothers fed standard diet (SD) and Fdmod (HLD) - offspring rats of diabetic mothers fed a hyperlipid diet. At 115 days of age, the OGTT will be performed again for glycemic evaluation and, at 150 days, the rats will be anesthetized and killed by decapitation to collect blood samples for oxidative stress biomarkers (thiobarbituric acid reactive substances and reduced thiol groups) in washed red blood cells. This project was approved by the Ethics Committee on Animal Use (Protocol No. 1218/2017). The data will be presented as mean and standard deviation and processed according to the most appropriate statistical tests. P <0.05 will be considered as the limit of statistical significance.
