Social relationship is a psychological and behavioral determinant throughout the life. Studies have showed that psychosocial stress may influence the cancer progression. However, there are no studies which have evaluated the influence of sociability behavior on chemically induced carcinogenesis. In this study, we will use an oral carcinogenesis model in rats to evaluate the effects of sociability behavior on the onset and progression of oral squamous cell carcinoma (OSCC) induced by 4-nitroquinoline-1-oxide (4NQO) carcinogen. Forty adult wistar mice will be submitted to sociability test and will be separated into two groups: group 1 - rats with higher sociable behavior and group 2 - rats with lower sociable behavior. After sociability behavior test, all animals will be treated with 4NQO carcinogen mixed in their drinking water during 120 days. After euthanasia, the extracted tongues will be submitted to histopathological examination to analyze the OSCC incidence, as well as to evaluate the tumor malignancy degree of both groups. Real-time PCR analysis will be performed to evaluate mRNA expression for OSCC progression related genes (VEGF, MMP-2, MMP-9, IL-6, TNF-alpha and p53). Specific statistical tests will be used to analyze the differences between the groups regarding OSCC incidence and growth, as well as tumor molecular profile. This study will allow us to understand possible connections between psychosocial and behavioral factors and the onset and progression of chemically induced cancer.
