Abstract

The aim of this study will be to investigate the anti-proteolytic potential against MMP-2 and MMP-9 and the ability to induce the remineralization of dentin through solutions containing different concentrations of TMP (in its cyclic form), different concentrations of F and association TMP/F. Root dentine blocks will be divided into 10 groups of experimental solutions (6mmx4mmx2mm, n = 10): 1) without F/TMP (Placebo); 2) 0.3% TMP; 3) 1% TMP; 4) 3% TMP; 5) 250 ppm F; 6) 500 ppm F; 7) 1100 ppm F; 8) 250 ppm F associated with 0.3% TMP; 9) 500 ppm F associated with 1% TMP and 10) 1100 ppm F associated with 3% TMP. For computerized microtomographic analysis, the selected root dentin blocks will be divided into 3 areas: 1-control-healthy area; 2-demineralized area - exposed to the demineralising solution for 7 days/37ºC, treated 3-demineralized area and subjected to pH cycling for 7 days at 37ºC. Next, the analysis of the mineral content of the root dentin (Micro-CT/gHAp×cm-3). will be done. The evaluation of the anti-proteolytic potential of the experimental solutions against the dentin matrix metalloproteinases (-2 and -9) will be carried out by zymographic analysis. For this analysis, the gels will be scanned and the intensity of the band formed by densitometry will be analyzed. For statistical analysis, if mineral content and zymography data present normal distribution and homogeneity, they will be submitted to ANOVA, followed by the Student-NewmanKeuls test. If the data are not homogeneous, the Kruskall-Wallis test will be used. It will be used from the statistical program Sigmaplot for Windows version 12.0 (SigmaPlot, Systat Software Incorporation, San Jose, CA), with significance at the 5% level. (AU)
