Antimicrobial photodynamic therapy using chlorin E6 in different concentrations and wavelength against single biofilm of periodontal pathogenic bacteria

Abstract

Scaling and root planing (SRP) is the treatment of choice for non-surgical management of periodontal disease. Nevertheless, this usual intervention might show limitations or insufficient outcomes when used as an unique therapy in certain clinical cases. Thus, alternative therapies have been investigated to overcome SRP deficiencies that may be related to the limited access to the root surface, presence of periodontopathogens in the deeper periodontal pockets and the ability of some bacteria to invade adjacent tissues. Among current alternative therapies, antimicrobial photodynamic therapy (aPDT) has been highlighted. This study aims at analysing the effect of aPDT using chlorin e6 in different concentrations and wavelength against single biofilm of bacteria related to periodontal disease. Single biofilms of Aggregatibacter actinomycetemcomitans will be formed, under apropriate conditions for 5 days, and treated with several protocols, including positive (chlorhexidine) and negative (distilled water + DMSO 1%) control, PS only, light only, and groups treated with aPDT (PS + Light). Different concentrations of chlorin e6, two wavelengths (blue and red) and one dose of energy (30 J/cm2) will be tested. Microbiological analysis will be carried out by counting colony-forming units (CFU/mL). Statistical analyzes will consider a significance level of 5% to compare the groups.
