Influence of bioglass functionalization, prepared by the sonochemistry route, with the 10% therapeutic drug in the cell response in vitro and in vivo

Abstract

With the increase in the life expectancy of the population, there was also an increase in the index of bone-metábolic diseases, with that it increased the search for strategies for regeneration and tissue repair bone. The objective of this study will be to evaluate the influence the association of the drug with bioglass in the differentiation and activity of mesenchymal cells and in the bone repair in critical defects performed in ovariectomized rats, aiming at orthopedic and odontological application. For the in vitro study, osteoblasts obtained from mesenchymal cells isolated from femurs ovariectomized rats. After differentiation, cells will be submitted to cell viability tests, total protein content, alkaline phosphatase activity (ALP), formation of mineralization nodules, measurement of cytokines and also the cellular interaction with the biomaterial will be evaluated. In the in vivo study, 48 Wistar rats will be used, which will initially be divided into two groups (n = 24), a bilateral varusectomy (OVX) group and a sham ovariectomy surgery group. After 30 days of these procedures, regardless of the group, all rats will be submitted to 5.0 mm critical bone defects on calvaria, which will be filled on the right side with clot (control) and on the left side half of the animals will receive (BV) and the other half will receive the 10% therapeutically biovidrofunctionalized (BVT) as the filling material. After 2 and 6 emanates, twelve animals from each OVX and Sham group will be euthanized, with n = 6 for each subgroup (BV and BVT), and the area of bone repair at calvaria will be assessed by histological, histomorphometric and immunohistochemistry. The data will be statistically analyzed, initially by means of the normality test aiming the application of the appropriate statistical test. The level of significance adopted will be the conventional value of 5%. (AU)
