Abstract

Obesity and pregnancy are factors in which systemic and oral changes can be noticed. The adipose tissue of obese patients secretes inflammatory mediators that exacerbate the inflammatory response. The mechanism during pregnancy is similar, due to estrogen and progesterone levels alterations. Therefore, both obesity and pregnancy are associated with high inflammation in periodontal tissues. Preliminary quantitative proton Nuclear Magnetic Resonance (1H-NMR) studies have demonstrated metabolic changes for multiple diseases, such as dental caries and periodontal disease. In addition, other studies have investigated effects of factors such as nutrition, dietary standardization and body mass index on salivary and plasma metabolites. The aim of this study will be to evaluate the systemic and periodontal conditions and the salivary and plasma metabolomic profiles of obese and eutrophic pregnant women. The sample will be consisted by 60 pregnant women divided into: obese (G1 = 30); eutrophic (G2 = 30) and they will be evaluated according to: a) contextual variables (household monthly income, schooling level and oral hygiene behaviors); b) anthropometric parameters; c) systemic condition (Gestational Diabetes Mellitus and Hypertension); d) Periodontal status (gingivitis and periodontitis); and e) 1H-NMR metabolic profile of saliva and plasma. Spectra will undergo principal component analysis, k-means cluster analysis and metabolite quantification. For comparison of contextual, anthropometric and periodontal variables, t test will be applied for quantitative variables with normal distribution and the Mann-Whitney test will be applied for assessment of the quantitative variables with non-normal distribution and ordinal qualitative variables. Finally, chi-square test will be used for nominal qualitative variables. The level of significance of 5% will be adopted. (AU)
