Proteomic profile of endodontic infections in diabetic patients

Abstract

Chronic endodontic infections are more prevalent in diabetic patients when compared to healthy patients, suggesting that Diabetes Mellitus may be a modifying factor for endodontic diseases. Endodontic infections are caused by a multi-species community of bacteria. As the methods of microbiological identification and quantification of endotoxins, are not able to infer the physiology, function and microbial pathogenicity, proteomic analysis is a technique that may contribute to the study of the pathogenesis of endodontic infections and the pathogen/host relationship. The present study aims: to qualitatively and quantitatively analyze the proteomic profile of endodontic infections in diabetic patients compared to systemically healthy patients; to correlate the expression of the bacterial proteins detected in the root canals and their biological functions in the two study groups. Samples will be collected from 18 patients referred for root canal treatment at the Faculty of Dentistry of Araçatuba FOA - UNESP, divided into two groups: control - systemically healthy patients (n = 9) and diabetic patients (n = 9). The sample will include asymptomatic teeth with primary endodontic infection, and presence of periapical lesion visible on radiography (chronic apical periodontitis). The identification of the peptides will be done in a nanoACQUITY UPLC-Xevo QTof MS system (Waters) using ProteinLynx Global Server (PLGS) software version 3.0, using the UniprotKB database (http://www.uniprot.org). The difference in protein expression will be statistically analyzed with Monte Carlo algorithm incorporated into the software (p <0.05). (AU)
