Analysis of bone metabolism under the influence of zoledronic acid and dexamethasone during socket healing process in C57BL/6J mice

Abstract

The homeostasis of bone metabolism can be influenced by a number of factors, including some medicaments, not only by those that directly interfere on bone cells, but also by those that act on the immunological system. This study aimed to evaluate the systemic effects caused by zoledronic acid (ZL) and dexamethasone (DX), separately, analyzing the serum levels of bone metabolism parameters during socket healing process in C57Bl/6J mice. A total of 30 male C57Bl/6J mice, ages of 12-18 weeks, mean weigh of 28 grams, will undergo upper right incisor extraction and divided in three distinct groups according to the treatment: Control - treated with 0.05 ml of 0.9% saline solution via IP, DX - 5mg/Kg of DX IP, and ZL - 0.5mg/Kg of ZL IP. The animals of all groups will be treated once a week for four weeks previously to the surgery, continuing until the euthanasia periods, of 7 and 30 days, when blood will be collected for alkaline phosphatase, total TRAP, calcium and phosphorous serum level analysis by means of colorimetric assays, as well as the maxillae with the dental sockets for histopathological analysis of the histological slices stained with hematoxilyn and eosin (HE), considering the presence of granulation tissue, maturation level of bone tissue, presence or not of bone sequestrum, microbial biofilm, and inflammation. Quantitative analysis will be performed after the capture of 10 fields of the medium region of the socket, stained with HE in a 100x magnification, and using a 130-point grid from ImageJ software. The points coincident with osteoblasts, osteoclasts (adhered or not), fibroblasts, osteocytes, and empty lacunae of osteocytes, bone matrix, bone sequestrum, collagen fibers, blood vessels, and leukocytes will be counted. The results will undergo normality distribution test for the selection of the adequate statistical tests, considering significant level of 5% (pd0,05).
