Influence of ultrasonic agitation on pH, biocompatibility and biomineralization of MTA Fillapex and Sealapex endodontic cements

Abstract

The purpose of this study is to compare the influence of ultrasonic agitation on pH, biocompatibility and biomineralization of MTA Fillapex and Sealapex cements. For the in vitro test, 50 polyethylene tubes will be prepared with one of the mouthpieces sealed with utility wax. The tubes will be filled with the tested cements and some will remain empty for control. The cements will be agitated with Ultra Sonic Piezoelectric Ultrasound for 20 seconds. After the setting time, the samples will be individually immersed in falcon tubes with deionized water and stored in a greenhouse. The pH will be measured in 10 minutes, 24 hours, and days 7, 15, 21 and 30. The solutions of each experimental time will be agitated and the pH will be measured and analyzed statistically. For the in vivo biocompatibility test, 40 male Wistar rats will receive subcutaneous implants of 4 polyethylene tubes containing the MTA Fillapex and Sealapex cements with and without agitation, and 1 tube for the control group. After the days 7, 15, 30 and 60, the animals will be euthanized and the polyethylene tubes will be removed with the surrounding tissues. After histological processing, the inflammatory infiltrate and fibrous capsule thickness will be histologically evaluated. The mineralization will be analyzed under polarized light. The data will be analyzed by the Kruskal-Wallis and Dunn test, with a significance level of 5%.
