Effect of chitosan on Candida species in salivary microcosm biofilms

Abstract

The present study aims to evaluate the effect of chitosan on Candida species in salivary microcosm biofilms developed on acrylic resin specimens. First, according to the microdilution method, it will be determined the minimum inhibitory concentration of chitosan capable of inhibiting the growth of Candida species in the planktonic state. A pool of saliva from five donors with oral candidiasis will be used as inoculum for formation of microcosm biofilms, which will be developed during 72 hours on acrylic surfaces using the Amsterdam Active Attachment model. After, biofilms will be treated for 24 hours with different concentrations of chitosan. Antibiofilm effect will be quantified by counting the number of cultivable cells, total biomass (crystal violet staining assay) and metabolic activity (XTT reduction assay). Biofilm structure after treatment with chitosan will be analyzed by confocal scanning laser microscopy. Appropriate controls will be included in all analyzes, and all tests will be performed in triplicate on three different occasions. Data will be analyzed to employ the most appropriate statistical test. P < .05 will be considered statistically significant.
