Abstract

Alzheimer's Disease (AD) is the most common cause of dementia in elders. Previous studies have shown as a possible mediator of the neurodegeneration an early-onset deficit in the metabolism of brain glucose and in the insulin signaling pathway. As a consequence, oxidative stress, neuroinflammation, activation of astrocytes and microglia, apoptosis, increase in the production of beta amyloid peptide (A²) and dysfunction of tau protein can occur. Nowadays, a key challenge related to AD is the development of new or prophylactic drugs. Bioactive compounds, derived or isolated from plants, have shown a potential neuroprotective effect in the treatment of neurodegenerative diseases. Thus, the goal of the study is to evaluate the potential neuroprotective effect of Eplingiella fruticosa Leaf Essential Oil Complexed With ²-Cyclodextrin (²-EF) in a pharmacological model of AD of sporadic type, induced by streptozotocin (STZ) in rats. Male Wistar rats (6-7 months old) will receive an injection of STZ (3 mg/kg, via intracerebral) and ²-EF (5 mg/kg, via oral daily). During the treatment, the following behavioral assays will be performed: sucrose preference, open field, olfactory sensibility, spontaneous alternation, and novel object recognition. In the end of the treatment, animals will be euthanized and their brains will be collected for the evaluation of protein expression by immunohistochemistry for A², IL-1² and IL-10 in different structures (olfactory bulb, hippocampus and cerebral cortex).
