Abstract

The purpose of the present study will be to evaluate the influence of oral CoQ10 supplementation on tibiae of rats modified systemically or not by nicotine. Eighty male rats (Wistar) will be divided into four groups (n = 20). In the previous thirty days, the animals will receive two daily subcutaneous injections of 3mg / kg of nicotine hemisulfate or saline solution in the dorsal region, with a 12-hour interval between From day 0, the protocol will constitute the administration via gastric gavage of a dose daily dose of 1ml of Coenzyme Q10 (CoQ10) or vegetable glycerin until euthanasia. SS (n = 20): also called the sham group. Animals belonging to this group will receive two daily subcutaneous applications of 0.5 ml of 0.9% sodium chloride (SS) solution throughout the experimental period. After 30 days after starting SS applications subcutaneously, the animals will start receiving daily, via gastric gavage, 1 ml of vegetable glycerin until the end of the experiment. SS-CoQ10 (n = 20): Animals belonging to this group will receive two subcutaneous injections daily with 0.5 ml of 0.9% saline solution, throughout the experimental period. After 30 days after starting SS applications subcutaneously, the animals will start receiving daily, via gastric gavage, daily supplementation with 120mg of CoQ10 until the end of the experiment. NIC (n = 20): Animals belonging to this group will receive two daily subcutaneous injections with 3 mg / kg of nicotine hemisulfate for a period of 30 days. After 30 days after the beginning of the applications of NIC by subcutaneous route, the animals will start receiving daily, via gastric gavage, 1 ml of vegetable glycerin until the end of the experiment. The animals will continue to receive subcutaneous applications of NIC for another 28 days until the end of the experiment. NIC-CoQ10 (n = 20): Animals belonging to this group will receive two subcutaneous injections with 3 mg / kg of nicotine hemisulfate for a period of 30 days. After 30 days after the beginning of the applications of NIC by subcutaneous route, the animals will start receiving daily, via gastric gavage, daily supplementation with 120 mg of CoQ10 until the end of the experiment. The animals will continue to receive subcutaneous applications of NIC for another 28 days. Euthanasia will take place at 7 and 28 days after the beginning of gavage. The collected pieces will be processed without demineralization for morphometric, densintometric, mechanical, micrtomographic analysis of the tibiae and as a complement, the histomorphometric analysis of the intestines. After analysis of normality and homoscedasticity, the data will be subjected to the most appropriate tests with a significance of 5% (p <0.05).
