Comparative study of the antitumor effect of latex from Janaúba (Synadenium grantii) in an in vitro model of breast carcinoma

Abstract

Breast tumors are the most frequently diagnosed neoplasms in female dogs, particularly those with an intact reproductive system. They represent about 50% of all tumors in females and almost 50% of the diagnoses of these tumors are found to be malignant. Despite the high frequency, there are few therapeutic options for breast cancer and few preclinical models for studies of this cancer. In this context, this research aims to establish a 3D culture model of canine mammary carcinoma cells and evaluate the antitumor effect of Janauba latex (Synadenium grantii) and doxorubicin, as a preclinical model for canine mammary carcinoma. Cell culture will be performed using three cultures of breast tumors. The cultured cells will be certified according to international guidelines. A cleavage assay of yellow tetrazolium salt will be performed to establish the inhibitory concentration of 50% of canine breast carcinoma cells. Subsequently, to analyze the antitumor effect of doxorubicin, Janauba latex and the association of doxorubicin and Janauba latex, the 2D culture will be tested with 4 different concentrations (0.5 ¼M, 1 ¼M, 2 ¼M and 2.5 ¼M). With this research we aim to find an antitumor effect or enhancement of doxorubicin with Janauba latex in the treatment of breast cancer cells, decreasing the dose necessary to inhibit cell proliferation and migration, bringing the hope of a new treatment to the veterinarian.
