Abstract

In vivo Fe-S cluster assembly into target apo-proteins requires a complex and conserved biosynthetic machinery. In non-green eukaryotes two systems are dedicated to mature Fe-S cluster containing proteins, known as ISC (Iron-Sulfur Cluster Assembly) and CIA (Cytosolic Iron-Sulfur Protein Assembly) systems. The ISC system takes place inside mitochondria and is responsible for the direct maturation of all Fe-S cluster containing proteins within the organelle and for the indirect maturation of all extra-mitochondrial Fe-S cluster containing proteins, through the export of an unknown sulfur-containing (X-S) compound. The CIA system occurs in the cytosol and rely on the X-S compound to mature all cytosolic and nuclear Fe-S cluster containing proteins. Our laboratory has identified an interaction between the CIA protein Dre2 and the R2TP cochaperone complex member Pih1/Nop17; moreover, other works showed the interaction between Hsp90 and other CIA proteins. The R2TP-Hsp90 complex matures a diverse range of client proteins, such as box C/D snoRNPs and RNA polymerase II. The interaction between either Nop17 or Hsp90 and CIA proteins suggest the participation of the R2TP-Hsp90 complex in the CIA system. Previous experiments in our laboratory have also indicated the presence of a [4Fe-4S] cluster in Nop17. In this project, we propose the characterization of the role of the R2TP-complex in the CIA system, as well as the structural characterization of the Fe-S cluster of Nop17. (AU)
