Proteomics study of hepatic tissues of broilers supplemented with seed maracujá oil

Abstract

Seed maracuja oil has been used in animal nutrition for presenting antioxidant substances and possibly promoting better production and animal welfare. The molecular mechanisms that the supplementation of seed maracuja oil can exert on the metabolism of animals can be studied as a function of changes in the proteome of fluids and/or biological tissues through proteomics. Thus, the objective of the research will be to investigate the effect of seed maracuja oil supplementation on productive performance, general metabolism, and biochemical profile and proteomic approach in broilers challenged under cyclic heat stress. The experiments with the animals will be conducted at the Poultry Nutrition Laboratory (UNESP/FMVZ /Botucatu), using one-day-old male chicks of the Cobb® 500 lineage and a completely randomized design. After the experimental period (42 days), the animals will be sacrificed and blood, liver and Pectoralis major muscle collected. In the proteomic study, the following strategies will be used: obtaining the proteomes by 2D PAGE, expression analysis of protein spots using Image Master Platinum v.7.0 software; Identification of differentially expressed protein spots by LC-ESI-MS/MS; functional analysis of proteins characterized in protein spots using Blast2GO, Cytoscape and DAVID softwares. Data will be analyzed using analysis of variance (ANOVA) and, when significant, means will be compared by Tukey test (P<0.05). For the inclusion levels of passion fruit seed oil, regression analysis will be applied using the SAS 9.4 ProcReg procedure.(AU)
