Evaluation of the influence of Coenzyme Q10 in the peri-implant repair process of implants installed in tibiae of rats exposed to nicotine

Abstract

The aim of this study will be to evaluate the influence of Coenzyme Q10 (CoQ10) on the peri-implant bone repair around implants in installed tibiae of rats systemically modified by nicotine. Forty male rats (Wistar) will be divided into two groups (n = 20). At day 0, the animals will receive an implant (4 x 2.2mm - SLA) in the proximal metaphysis of the right and left tibiae. 30 days before and 28 days after surgery, the animals will receive two daily subcutaneous injections of 3mg/kg of nicotine hemissulfate (NIC) in the dorsal region (12/12 h). Right after surgery, the protocol will consist of daily supplementation with either 1 ml of vegetable glycerin or 120 mg of CoQ10, via gastric gavage, both until the end of the experiment. Group NIC: the animals will be subject to NIC and supplementation with 1 ml of vegetable glycerin. NIC-CoQ10: the animals will be subject to NIC and supplementation with 120 mg of CoQ10. Animals will be euthanized at 7 and 8 30 days after surgery. The collected pieces will be processed with demineralization for histological analysis, area of newly formed bone tissue and immunohistochemistry for detection of TRAP and without demineralization for bone-implant contact analyses. After the analysis of normality and homoscedasticity, the data will be discovered using the most appropriate tests with a significance level of 5% (pd0.05).
