Abstract

This proposal has as main objective the development of microfluidic devices to evaluate the quality of bovine embryos. The aim is to dynamically observe the embryonic process and the quality of the embryos. To assess the quality of the embryos, the culture medium will be collected for 3 days and the extracellular vesicles (EVs) present in this medium will be characterized. The quality of embryos in microdevices and in the conventional method (macroscale) will be estimated by analyzing sizes, loads, concentration of miRNA and DNA present in the EVs. For this, the microdevices will first be operationally and biologically validated (biocompatibility), concomitantly extracting the oocytes and characterizing them. After that, the oocytes will be fertilized (embryo formation) in microdevices and in plates for 3 days. During this period, samples will be taken from the culture medium and the EVs resulting from the embryonic process will be characterized. Through the characteristics of these EVs, it will be possible to determine good quality embryos. The results obtained in microdevices will be statistically analyzed and compared to the conventional system (macroscale).
