Analysis of the osteoinductive principle of bioactive glass incorporated into the surface of miniplates by the anodizing technique: study in diabetic rats

Abstract

The present study aims to evaluate the osteoinductive capacity of a new bioactive glass-based coating synthesized via electrolytic plasma oxidation (PEO-BG) in bone neoformation in diabetic rats. For this, commercially pure titanium (Ti-cp) miniplates will be coated by PEO-BG (~ 45.0 Si, 24.5 Ca, 24.5 Na, 6.0 P; m/v %) via PEO (500 V, 1000 Hz, duty cycle of 10%, 420 s).Initially, surface characterizations will be conducted to confirm the morphology (Scanning Electron Microscopy - SEM), topography (Confocal Laser Scanning Microscopy - CLSM), and chemical composition (Energy Dispersive Spectroscopy - EDS; X-Ray Excited Photoelectron Spectroscopy). - XPS) and physicochemical properties (Profilometry, Wettability, and Surface Free Energy) of the samples. Then, the in vivo study will be conducted using Wistar rats (n=12) and a protocol of induction of diabetes mellitus by the administration of streptozotocin (35 mg/kg) will be used. After 30 days of diabetes induction, the animals will be randomized and allocated to the miniplate analysis (n=12). The rats will undergo fracture and osteosynthesis surgery, the other animals will be randomly fixed to the test miniplates (PEO-BG) and conventional miniplate (CONV) in each femur. The animals will be euthanized and the pieces sent for analysis of the calcified tissues (28 days). Calcified tissues will be analyzed by Computerized Microtomography (Micro-CT) and CLSM scanning to assess bone tissue dynamics, as well as histometric analysis to determine the area of newly formed bone and osteoinductive/osteoconductive effectiveness of the coating. The quantitative results will be submitted to the normality test (Shapiro-Wilk) and a parametric or non-parametric test will be applied for p<0.05.(AU)
