Abstract

The intense use of pesticides in agricultural areas around the world exposes serious risks to the environment and human beings. The identification of water and food contaminated with penconazole shows that exposure of non-target species occurs and is of great concern. In vitro metabolism models can be used to evaluate pesticide metabolism. This technique has several advantages such as the simplicity of carrying out the experiments, low amount of substrate needed for the studies, and, mainly, the lower or absence of animals. Among the various models used to study in vitro metabolism, human liver microsomes is the main model used, as it is rich in cytochrome P450 enzymes. The penconazole molecule has an asymmetric center; thus, the enantioselective evaluation of metabolism in vitro in human models may provide new information about this pesticide, since penconazole has already shown stereoselective behavior in studies involving other organisms and an enantioselective study has not yet been performed in human models. Thus, this project aims to validate an enantioselective method for the analysis of the chiral pesticide penconazole in human liver microsomes and a subsequent pilot study of in vitro metabolism. The data obtained will serve as subsidies for the determination of enzymatic parameters and subsequent in vitro-in vivo correlation. (AU)
