Verification of the presence of EcPV 8 and 9 DNA in equine aural plaques from Brazil

Abstract

The Equus caballus papillomavirus (EcPV) has been associated with some neoplasia in horses, e.g., classical papillomatosis, genital papilloma, and aural plaque. Aural plaque is a cosmopolitan dermatophyte characterized by the presence of circular, whitish lesions up to 3 cm in diameter in the inner surfaces of the pinna and rarely in other regions of the horse's body, regardless of age, breed, or gender. The EcPV1, 3, 4, 5, and 6 have previously been associated, alone or in coinfection, with an equine aural plaque in Brazil, whereas, EcPV2 and 7 were not identified in these samples. However, due to the recent discovery of EcPV8 and 9, no studies have evaluated the presence of these EcPVs in equine aural plaque samples. Therefore, the aim of this study will be to verify the presence of EcPV8 and 9 DNA by PCR in 83 samples of aural plaque from horses from 30 farms located in different regions of Brazil. These samples were used in a previous study that evaluated the presence of EcPV1 - 7. The specific primers for EcPV8 and 9 will be synthesized and tested in silico. The PCRs specificities will also be proven by direct Sanger sequencing of positive PCR products. As the association of EcPV8 and 9 to the aural plaque is unknown, mini-genes will be built that will serve as positive PCR controls. Furthermore, the clinical characteristics of the aural plaque will be evaluated according to the EcPV detected. At the end of this experiment, we seek to assess the importance of these viruses in the etiology of the aural plaque and, if possible, associate them with the clinical alterations of this dermatopathy.(AU)
