Abstract

The aim of these studies is to analyze the differences in susceptibility to periodontal disease (PD) after systemic microbial challenge in known susceptible (Balb/C) and resistant (C57BL/6) mouse strains. For the first and second objective, 140 mice were used, divided into 2 groups (n=70 Balb/C & n=70 C57BL/6). Each group was divided into 3 subgroups: Control (C), Sham vehicle with 2% sterile carboxymethylcellulose (Sham), Porphyromonas gingivalis W83 Gavage (Gav). The microbial challenge was performed weekly for 3 weeks, and after this period, the mice were kept until completing 42 days of the experiment, when they were then euthanized. The following have already been evaluated: 1) alveolar bone microarchitecture in the region of the upper 2nd molar using x-ray computed microtomography (Micro-CT); The following are being evaluated: 2) severity of inflammation present in the interproximal region of the maxillary second molar through histomorphometric analysis; 3) expression of epigenetic methylation markers; (DNMT3b) and histone acetyl histone 3 (H3) and acetyl histone 4 (H4) acetylation through indirect immunofluorescence in the intestine and maxilla; The Methylation Array has already been carried out and now is read for bioinformatics (analysis proposed for conducting the Research Internships Abroad (BEPE), for one year). The data obtained will be submitted to statistical analysis (p < 0.05). (AU)
