Identification of biomarkers from blood serum extracellular vesicles in bitches with mammary tumors.

Abstract

Canine mammary tumors are responsible for a large case number in the small animals' clinical routine, being one of the neoplasms that most affect bitches. Breast cancer in women is considered common cancer, with high morbidity and mortality, similar to veterinary medicine. The extracellular vesicles (EVs) from blood serum have been studied to identify biomarkers for early diagnosis, prognosis, monitoring, and treatment of several types of cancer. EVs have specific functions in oncology, such as tumor growth and progression, modulation of the tumor microenvironment, and delivery of pro-metastatic information. Thus, this study aims to evaluate EVs proteomics from the blood serum of bitches affected or not by CMT and correlate these proteins with the histopathological malignancy grading of these tumors. A total of 30 bitches from the routine of the Animal Reproduction service, Veterinary Hospital, FMVZ, UNESP de Botucatu will be used. The animals will be into three groups, control (young bitches, no evidence of breast cancer, n = 10), less invasive breast tumor (n = 10), and more invasive breast tumor (n = 10), according to the tumor staging and degree of malignancy. History, anamnesis, physical exam, and case evolution will be assed at the animal's first clinical evaluation. The canine mammary tumos will be evaluated for macro (pre-surgery) and microscopic (pre [aspiration cytology] and post-surgery [histopathology]) characteristics. During surgery, blood samples to separate the EVs by centrifugation and a commercial kit will be collected. The isolation of EVs will be confirmed by transmission electron microscopy. EVs will be processed, and proteins extracted for mass spectrometry (MS). MS results will be evaluated by a multivariate analysis using PCA, PLS-DA, and VIP scores to compare groups. The expectation is to identify proteins correlated with the disease's malignancy
