EVALUATION OF THE REDOX STATUS OF SUBMANDIBULAR GLANDS OF WISTAR RATS WITH INDUCED APICAL PERIODONTITIS

Abstract

The aim of the study will be to evaluate the redox status of the submandibular glands of Wistar rats with induced apical periodontitis. For that, 24 rats will be distributed in two groups (n=12): control group (C) and group where apical periodontitis (AP) will be induced in the first and second right molars of the maxilla and mandible. After 28 days, the rats will be euthanized and then the right and left submandibular glands will be removed, weighed and stored at -80°C. The right hemi-maxilla and hemi-mandible will be removed and histologically processed to confirm the periapical lesions. The submandibular glands on both sides will be processed concomitantly for biochemical analysis to compare possible differences due to the fact that periapical lesions are performed unilaterally. For biochemical analyses, supernatants from submandibular gland homogenates will be used for spectrophotometric assays: a) total protein content; b) lipid oxidative damage will be determined by the thiobarbituric acid reactive substances method; c) the protein oxidative damage will be determined by the carbonylated protein method; d) total non-enzymatic antioxidant capacity will be evaluated by the antioxidant power of reducing total ferric, reduced glutathione and uric acid; e) enzymatic antioxidant capacity will be determined by the activity of the enzymes superoxide dismutase, catalase and glutathione peroxidase. The normality and homoscedasticity of the data will be analyzed and they will be submitted to the most appropriate parametric or non-parametric test. For all tests, the level of rejection of the null hypothesis will be set at 5% (p < 0.05).
