Abstract

This project aims to perform field collections and to obtain chromosomes from the species Lepidosiren paradoxa (Sarcopterygii; Dipnoi), popularly known as piramboia. To date, Dipnoi represent one of the most challenging groups for obtaining chromosomes. Despite representing one of the ancestral groups of vertebrates of greatest evolutionary interest, available chromosome studies are limited to a study carried out in the 1960s that indicates the presence of 2n=38 chromosomes, where only illustrations of their chromosomes are represented. Given our current accumulated expertise in obtaining chromosomes in all groups of vertebrates using different methodologies, we will seek to overcome these methodological challenges and obtain chromosomes from piramboia using multiple approaches, such as lymphocyte cell cultures, fibroblast cultures, fin regeneration, and conventional direct preparations (from kidney or spleen cell cultures). Recently, the piramboia genome was assembled (NCBI GCA_040581445.1), indicating the presence of 38 linkage groups, evidencing that it is the largest genome of all animals analyzed to date (with approximately 91 GB). One of the proposed future objectives is to anchor these genomic sequences to the chromosomes of the species through the use of synthetic oligonucleotide-based probes.
