Abstract

Plant cell walls are an important renewable raw material for the sustainable generation of fuels, materials and chemicals. The recalcitrance of the complex network of polysaccharides and lignin that make up the lignocellulosic component of plant cell walls makes it difficult to deconstruct the biomass by hydrolytic enzymes. Detailed knowledge of cell wall architecture can provide a rationale for improving enzymatic degradation by targeting specific polysaccharides. As a proof of principle, we propose to study the effect of enzymatic treatment of plant cell walls using cell surface sensitive techniques to follow the effect of enzymatic activity on cell wall architecture. The effect of hydrolytic enzyme treatments on polysaccharide content and distribution in sugarcane stem sections will be evaluated by monitoring the binding of extrinsically labeled carbohydrate binding modules using images generated by fluorescence lifetime imaging microscopy (FLIM) along with advanced secondary ion mass spectrometry (SIMS) based imaging.
