Influence of doxycycline on the expression of inflammatory genes in dental alveolus of osteopenic rats

Abstract

The tooth extraction is a common dental problem and can be due to trauma, decay, periodontal diseases or prosthetic indication. Patients with osteoporosis have alterations in the bone metabolism and they need a longer treatment time to permit the bone repair when this patient go on rehabilitation with implants or orthodontic treatment. Furthermore, a correlation between osteoporosis and bone fracture is because the less density of the bone in these patients. The aim of this study is compared, in Wistar rats, the inflammation and repair after tooth extraction using doxycycline at sub antimicrobial doses (SDD) and alendronate. In this study, 36 Wistar rats will be used, divided in 6 groups: C - Control; CDOX - Control exposed to doxycycline 30 mg/kg/day; CAL - Control exposed to alendronate sodium 0.7 mg/kg/week; OVX - Control Ovariectomized; OVXDOX - Ovariectomized exposed to doxycycline 30 mg/kg/day; OVXAL - Ovariectomized exposed to alendronate sodium 0.7 mgkg/week. The rats, with about 80 days of age, will undergo the osteopenia induction procedure and it is known that in 90 days it will be established. Afterwards, the rats will start drugs treatments with a dose of alendronate sodium 0,7 mg/kg/week or at a dose of doxycycline 30 mg/kg/day, both for 60 days. The total experimental period will be 150 days, and three and five days before the euthanasia, all animals will have tooth extraction procedure. After 150 days from the beginning of the experiment, the animals will be euthanized. The samples collected will be analyzed by Real-time qRT-PCR. The results of this project will be selected to the multivariate linear model test.
