Abstract

This study aims to evaluate the effects of incorporating the antibacterial monomer DMAHDM (2.5% or 5%) with or without NACP (2%) on the antibacterial potential and remineralizing potential of an experimental resinous infiltrant. Real-time metabolic status of non-disrupted Streptococcus mutans biofilms (UA 159 Luc-ldh) will be assessed using a high-throuput bioluminescence assay (n=18/group), the 3-dimensional distribution of biofilm components (nucleic acids, proteins and EPS) using a concurrent staining protocol and confocal microscopy analysis (n=3 specimens/group; 3 locations/specimen), and the quantification of the total biovolume (¼m3), total number and percentage of live cells of the bacterial biofilm of Streptococcus mutans through the Live/Dead assay (n=12 specimens/group, 3 locations/specimen). Bioactivity analysis will also be performed through Scanning Electron Microscope and X-ray diffraction analysis (n=12). Data will be analyzed for normality and homoscedasticity, and statistical analysis will be performed with appropriate tests with a significance level of 5%. (AU)
